RESUMO

The two-component system (TCS) genes are involved in a wide range of physiological processes in prokaryotes and eukaryotes. In plants, the TCS elements help in a variety of functions, including cell proliferation, response to abiotic and biotic stresses, leaf senescence, nutritional signaling, and division of chloroplasts. Three different kinds of proteins make up the TCS system in plants. These are known as HKs (histidine kinases), HPs (histidine phosphotransfer), and RRs (response regulators). We investigated the genome of Gossypium raimondii and discovered a total of 59 GrTCS candidates, which include 23 members of the HK family, 8 members of the HP family, and 28 members of the RR family. RR candidates are further classified as type-A (6 members), type-B (11 members), type-C (2 members), and pseudo-RRs (9 members). The GrTCS genes were analyzed in comparison with the TCS components of other plant species such as Arabidopsis thaliana, Cicer arietinum, Sorghum bicolor, Glycine max, and Oryza sativa. This analysis revealed both conservation and changes in their structures. We identified 5 pairs of GrTCS syntenic homologs in the G. raimondii genome. All 59 TCS genes in G. raimondii are located on all thirteen chromosomes. The GrTCS promoter regions have several cis-regulatory elements, which function as switches and respond to a wide variety of abiotic stresses. RNA-seq and real-time qPCR analysis showed that the majority of GrTCS genes are differentially regulated in response to salt and cold stress. 3D structures of GrTCS proteins were predicted to reveal the specific function. GrTCSs were docked with abscisic acid to assess their binding interactions. This research establishes the groundwork for future functional studies of TCS elements in G. raimondii, which will further focus on stress resistance and overall development.

Rhizosphere acidification in leguminous plants can release P from the dissolution of phosphate compounds which can reduce Pb bioavailability to them via the formation of insoluble Pb compounds in their rhizosphere. A soil polluted from Pb-acid batteries effluent (SPBE), having total Pb = 639 mg kg-1, was amended with six different rates (0, 0.5, 1, 2, 4 and 6%) of oxalic acid-activated phosphate rock (OAPR) and their effects on pH, available P and bioavailable Pb concentrations in the rhizosphere and bulk soils of mung bean plant were evaluated. Furthermore, the effects of these variant OAPR rates on Pb concentrations in plant parts, bioaccumulation factor (BAF) and translocation factor (TF) for Pb in grain and traits like productivity, the activities of antioxidant enzymes, and grain biochemistry were investigated. Results revealed that increasing rates of OAPR significantly increased pH values and available P while decreased bioavailable Pb concentrations in the rhizosphere over control. The highest dissolution of P in the rhizosphere was with 4 and 6% OAPR rates. As a result, the formation of insoluble Pb compounds affected on reduced Pb concentrations in shoots, roots, and grain in addition to lower grain BAF and TF values for Pb over control. Likewise, the highest plant productivity, improved grain biochemistry, high Ca and Mg concentrations, least oxidative stress, and enhanced soil alkaline phosphatase activity were found with 4 and 6% OAPR rates. The OAPR 4% rate is suggested for reducing grain Pb concentration, cell oxidative injury, and improving grain biochemistry in mung bean.

Soil pollution with Cd has promoted serious concerns for medicinal plant quality. Amending Cd-polluted soils with textile waste biochar (TWB) coated with natural polymers can lower Cd bioavailability in them and reduce associated environmental and human health risks. In this study, we explored the impacts of solely applied TWB, chitosan (CH), their mix (TWB + CH) and TWB coated with CH (TBC) in Cd-polluted soil on Cd distribution in moringa (Moringa oleifera L.) shoots and roots as well as plant-available Cd in soil. Moreover, amendments effects on plant growth, dietary quality, and antioxidative defense responses were also assessed. Results revealed that the addition of TWB, CH, and TWB + CH in Cd-polluted soil reduced Cd distribution in shoots (56%, 66%, and 63%), roots (41%, 48%, and 45%), and plant-available Cd in soil (38%, 52%, and 49%), compared to control. Interestingly, the TBC showed significantly the topmost response for reducing Cd concentrations in shoots, roots, and soil by 73%, 54%, and 58%, respectively, relative to control. Moreover, amending Cd-polluted soil with TWB, CH, and TWB + CH depicted significantly better effects on plant growth, dietary quality, and activities of soil enzymes but the topmost response was observed with TBC treatment. Compared with control, TBC improved plant growth parameters: shoot length (81%), root length (90%), shoot fresh weight (60%), root fresh weight (76%), shoot dry weight (75%), root dry weight (68%) contents of chlorophyll-a (42%) and chlorophyll-b (74%), and soil enzyme activities: urease (130%), catalase (138%), protease (71%), cellobiohydrolase (45%), acid phosphatase (34%), peroxidase (60%), ß-glucosidase (152%), chitinase (62%), and phosphomonoesterase (139%). Furthermore, TBC treatment arrested Cd-induced oxidative stress via escalating the activities of antioxidant enzymes as well as improved moringa dietary parameters (protein, tannins, lipids, alkaloids, saponins, terpenoids, flavonoids, and tocopherols contents). Such findings suggest that the TBC has an immense perspective to remediate Cd-polluted soils and prevent human health risks associated with Cd exposure through the diet.

Past studies have thoroughly explored the phytoextraction/phytostabilization potentials of different plant species for particular metals. However, none of the plants was able to tackle the problem of multi-metal in contaminated soils. We report herewith the potential of Conocarpus erectus to extract lead (Pb) while having the capability to stabilize chromium (Cr), nickel (Ni) and cadmium (Cd) in polluted soil. The C. erectus was subjected to grow for 120 days in a soil spiked with four different levels of each metal i.e. Pb (0, 600, 1200 and 2400 mg kg-1), Ni (0, 50, 100 and 200 mg kg-1), Cr (0, 150, 300 and 600 mg kg-1) and Cd (0, 20, 40 and 80 mg kg-1). Data related to plant growth, physiology, biochemistry and antioxidants activities revealed that forenamed parameters were significantly reduced with increasing spiking levels. Contrarily, metal speciation in plant parts (metal concentrations in shoots and roots, and metal contents in these corresponding plant parts), metal removal per pot, and DTPA-extractable metals from the soil were significantly increased with increasing spiking level upon the termination of the experiment. Curiously, each spiking level demonstrated elevated Pb concentrations in shoots than roots, while the concentrations of other metals (Cr, Ni, and Cd) were found higher in roots than in the shoots. Likewise, at each spiking level, C. erectus showed both bioconcentration factor (BCF) and translocation factor (TF) values greater than 1 for Pb, while these values were ever lower than 1 for Cr, Ni, and Cd. Moreover, the percentages of Pb removal were ever higher than other metals at each spiking level. Outcomes of our experiment suggest that C. erectus has immense potential for the phytoextraction of Pb and phytostabilization of Cr, Ni, and Cd in polluted soil. It is suggested that this plant can be used to tackle the problem of multi-metal pollution in soils.

Diabetes mellitus is a life threatening disease and scientists are doing their best to find a cost effective and permanent treatment of this malady. The recent trend is to control the disease by target base inhibiting of enzymes or proteins. Secreted frizzled-related protein 4 (SFRP4) is found to cause five times more risk of diabetes when expressed above average levels. This study was therefore designed to analyze the SFRP4 and to find its potential inhibitors. SFRP4 was analyzed by bio-informatics tools of sequence tool and structure tool. A total of three potential inhibitors of SFRP4 were found, namely cyclothiazide, clopamide and perindopril. These inhibitors showed significant interactions with SFRP4 as compared to other inhibitors as well as control (acetohexamide). The findings suggest the possible treatment of diabetes mellitus type 2 by inhibiting the SFRP4 using the inhibitors cyclothiazide, clopamide and perindopril.

OBJECTIVE: To determine the frequency of microbiological contamination of water in different water filtration plants in Islamabad. STUDY DESIGN: Descriptive cross-sectional study. PLACE AND DURATION OF STUDY: Water Filtration Plants (WFP) in different sectors of Islamabad, from April to September 2012. METHODOLOGY: Water samples were collected in sterilized bottles according to the standard water sampling protocol from site and transported to Pakistan Council for Research in Water Resources (PCRWR) for analysis. Microbiological quality of water was determined in terms of total coliforms (< 2.0 MPN/100 ml) and Escherichia coli (< 2.0 MPN/100 ml). Microbiological contaminated water was defined the sample which had more than 2.0 MPN per 100 ml of either total coliforms or Escherichia (E.) coli. RESULTS: Thirty two WFP were analyzed for microbiological contamination. E. coli was present in 8 (25.0%) water samples, while 24 (75.0%) water samples were free from it. Total coliforms were present in 13 (40.6%) of the samples of WFP, while 19 (59.3%) samples were free from total coliform. Faecal coliforms were present in 8 (25.0%) and absent in 24 (75.0%) samples. Both E. coli and total coliform were present in 8 (25.0%) samples. Nine (59.3) WFP were free from E. coli, total coliform and faecal coliform. Statistically, no significant association was found (p > 0.05) between microbiological contamination and the sectors. CONCLUSION: Less than half of the water samples of the WFP were contaminated while certain sectors showed more frequent contamination than others.

RESUMO

Non Hodgkin's Lymphoma (NHL) comprises a group of lymphoproliferative disorders the frequency of which continues to rise. Although many classification systems exist for identifying specific histological subtypes, NHL is generally divided into indolent (low-grade) and aggressive (intermediate- and high-grade) forms. Large B Cell Lymphoma (LBCL) is one of the commonest aggressive NHLs. The aim of this review is to provide a general overview of NHL, its clinically practicable cellular classification, epidemiology and in depth overview of the evolution of treatment of LBCL during the past 5 years. Current guidelines from National Cancer Institute (NCI), USA and National Institute on Clinical Excellence (NICE), UK are mentioned and recommendations according to our own set-up are suggested.
